The main character Lucky Luke is a lonely poor cowboy of the western United States in the late 19th century. With the nickname "shoot faster than his shadow," Lucky Luke wanders the West to protect justice and right.

In the first episodes, Lucky Luke's appearance and personality were not really shaped. In later stories, two authors, Morris and René Goscinny, built the image of Lucky Luke as a funny, single, plain cowboy with a white hat, black vest, yellow shirt and neck. wear a red scarf. At first, Lucky Luke always appeared with a cigarette on his lips, but later to avoid the image of a smoking hero, Morris replaced it with a blade of grass. Handling situations intelligently, plus shooting skills and luck, Lucky Luke always succeeds at the end of each volume.

Along with Lucky Luke is Jolly Jumper, the fastest horse in the West. As a special horse, Jolly Jumper is walking on the wire, so smart that he plays chess with Lucky Luke and when talking the story can lead to literature. Jolly Jumper repeatedly saved Lucky Luke from difficult situations but these two characters often teased each other. As in the episode of Sarah Bernhardt, Jolly Jumper disparaged because Lucky Luke looked in the mirror before going to see Sarah Bernhardt, while Lucky Luke teased that Jolly Jumper wore nothing when the horse was not wearing a saddle. Jolly Jumper in particular hates the Rantanplan dog.

The most frequent villain in the comics is the Dalton brothers. From models of real characters in Western history, Morris built four Dalton bandits but killed them as soon as they appeared. Later, Morris and René Goscinny resurrected them by appearing to Dalton's cousins.

The following Dalton bandits include: Joe, Jack, William and Averell. Joe is the oldest, shortest but also the most aggressive, the leader of the group. Then Jack, William, gradually higher but also less aggressive. Finally, Averell, the youngest, tallest and most stupid family. The volumes usually begin with the Dalton brothers fleeing from prison, and end when they are captured by Lucky Luke.
